IMDB has page on a 2001 film of this name but it may not be the same film as the one whose info is given here (the youtube page given below says the film was released in 2004 and the hinduonnet.com page lists a different director to the IMDB page).

According to the hinduonnet.com page: Uday Kiran "plays Gautam who comes to Hyderabad to join a computer institute. ... Gautam and his friend Bablu move into a house owned by one Ramana, as paying guests. Gautam meets Anju (Venya) and teams up with her at the computer institute to do a programme. And their pair is called `Jodie No.1' While Gautam falls in love with Anju, she remains indifferent. Meanwhile, Gautam finds out that Anju's pen friend Pinky is planning to visit her. His well-wisher and landlord Ramana asks him to grab this opportunity. Thus Gautam arrives in a female garb impersonating as Pinky."
The youtube page for the clip adds this: "Jodi no:1 was made in telugu and released in 2004. I played a supporting role in which I fall in love with the wrong person who turns out to be a man in the end." The name of the person who posted the clip is given on the youtube profile page: "Ernst Thomas, Actor-Writer-Executive Producer-Director".
